There's a connector at the gearbox.
There's a connector at the rear lights
& there's a connection at the fuse box, but that one may be a general live feed, so may not be helpful to you.?

A few things to check would be the coolant temp sender, TPS sensor and last but not least check for a vacuum leak. These are just a couple things off the top of my head.

Try stepping on the brake when trying to shift with the key on. Then try starting im neutral or other gears to see if the neutral safety switch is bad in park. You may have a bad wire from the battery to power distrubution or bad ground. So many things it could be.

most relays are located in fuse box, some cars have more than 1 fuse box in them.

there should be a panel on the inside side of the tailgate with screws, remove panel to gain access to the latch assembly u have a rode off or bent

Your vehicle has a PCB - relay . Printed circuit board relay . Soldered directly on the circuit board inside the under hood fuse box . You could unsolder the the relay an solder in a new one , how are your solder skills ? Your could also buy a regular relay an socket an find a wiring diagram so you know what wires to cut an splice the new relay socket too.

The 2010 GMC Sierra does not have a separate flasher. The turn signal function is built into the body control module. You cannot replace the flasher alone. If you have a scan tool, you can connect to the DLC and see if the turn signal switch command is being received by the BCM.

This scenario could be something as simple as just the exhaust vibrating at a certain time or engine speed and vehicle speed not matching at a point and then the exhaust vibrates against the underside of the floorboard because it is so close to the floor.
