ahhh the mayhem... it may very well be the curse of Vista, Which doesn't play well with CAT software or interfaces been there done that... Windows 7 or XP are the best bets, then you need to ensure your settings all match, and do NOT set your connect rate faster than 9600 (baud rate) since there really is no reason to go faster, and if you do it can cause issues. make sure (can not stress this enough) that settings match...

My FT2000 has the last Yaesu´s updated firmware and also the AC0C roofing filter installed in. The FT2000 with those changes seems another class of radio.

The problem is likely the TX/RX relay. If it is old (and I am sure it is) the contacts likely need to be cleaned, or the relay replaced. It is a common enough relay if you want to replace it. Also, pull each PCB card and clean the contacts with a pencil eraser. Pink erasers are OK but the white ones are better as they aren't as abrasive.

Hope this helps, VE3CMQ

a couple things you can try if nothing shows on the screen at all.
Do a full reset first of all, that's the best step with any hand held radio problems.
Try pressing F then 0 and if that don't work try battery removal and if you still get nothing time to call Yeasu they have always had firmware issues with handhelds I have a triband handheld that did the same thing

Check your transmit frequency to make sure it is within the proper range. Also verify proper battery voltage, it will typically transmit with low voltage.

I am not sure, but this usually indicates that the PLL has lost lock. These days, it could be as simple as a poorly soldered connection. I don't recommend that you try repairing this yourself as there are essentially no user serviceable parts in radios these days.

Radio repairs these days have come down to board swapping because the parts are so tiny that humans have a hard time replacing (and in some cases, seeing) the parts. Yaesu US has a very good reputation for quality repairs so I'd suggest you send it there. They will likely just swap a board out, but that will be less expensive than having someone spending 10 hours trying to find the problem and repairing it. I know, as I work in a radio shop, and if it isn't something real simple, all we can do is do a board swap.

Check the fuses and the power supply. Make sure that the power supply has enough capacity to run the radio, at least 20A! Make sure that the power supply is putting out 13.6V.

If you were unfortunate enough to hook the power leads up backwards, then a trace may be blown off the circuit board internal to the radio. Also, it is likely that the reverse polarity diode inside has shorted out. This may cause the fuse to blow when you apply power. If it has blown, it likely took the the trace out inside the radio.

If the power supply and fuses are OK, unless you are competent technically, the only recourse is to take the radio into a shop and get them to fix it. If you were upgrading the firmware inside the radio when it happened, then the radio is likely non-repairable.

I don't recommend ever upgrading a radio's firmware! If it isn't broken, then don't fix it!!

My guess is that the power supply you are using is not up to the task. You need one that will provide at least 15 amps at full output. Try setting your radio to a lower power setting and see what happens. You may find that you have to upgrade your power supply.

It might be the RF switching diodes in the receive antenna circuitry. These sometimes get damaged due to lightning strikes nearby or hi SWR levels. That would be my first guess.

It sounds to me that your car is generating noise on the power lines and causing noise on the transmit signal. Make sure that your positive and negative leads from the radio go directly to the cars battery! This will help prevent this problem.

Unplug the mic. If it stays keyed up, then you have a problem within the radio. If it un-keys, then the problem is in the microphone. If it is internal to the radio, take it to a competent technician. If the problem goes away when the mic is unplugged, then get a new microphone.
